PHYTOCHEMICAL CHARACTERIZATION AND EVALUATION OF QUERCETIN FOR ANTI-ARTHRITIS FROM ANNONA SQUAMOSA ROOT

Abstract

Rheumatoid arthritis is an immune mediated inflammatory disorder. The immune system affects the tissues of the joints this is due to discharge of certain chemicals and enzymes that eat up the bones and cartilage. Rheumatoid arthritis mainly affects women than men (3:1) and generally, it strikes between the age of 30 and 55 years and also it affects 0.5-1.0% of adults. Annona squamosa is a medicinal plant which is used for the treatment of arthritis in Ethnopharmacology. The present study is aimed to investigate the therapeutic activity of ethanolic extract from Annona squamosa root. The researcher was undergone to isolate and purify the ethanolic extract from the root of Annona squamosa by Column chromatography. Further the purified bioactive flavonoid was characterized by TLC, UV, H1 NMR, C13 NMR and HPLC and found to be 2-(3, 4-dihydroxyphenyl)-3, 5, 7 - trihydroxy-4H-chromen-4-one. The isolated flavonoid compound was tested for Anti-arthritic activity, which showed the inhibition for HRBC membrane stabilization (66.37%) as compared with standard Diclofenac sodium (79.31%) at a concentration of 100?g/ml. Anti-arthritic activity of isolated compound showed the inhibition for egg albumin (65.17%) as compared with standard Diclofenac sodium (78.57%). This research proved that the isolated compound 2-(3, 4-dihydroxyphenyl)-3, 5, 7 - trihydroxy-4H-chromen-4-one which is Quercetin, a flavonoid from the ethanolic extract of Annona squamosa root have anti- arthritic activity.
